  • Finding Genes. The National Center for Biotechnology (NCBI)  provides many tools for identifying sequence.
    • Genbank  Input words such as "tryptophanase" and "Escherichia" to find sequence of interest.
    • BLAST  Use BLAST if you know some sequence, and want to find other similar sequences, either in E. coli or other organisms.
    • Kenn Rudd's Ecogene  Find your gene on the map, along with its sequence, and possible regulators.
    • Ecocyc for metabolic pathways
  • Analyzing DNA
  • Primer design for PCR.
    • Primer3 Primer design and analysis program.  Once you know which gene regulatory region to amplify, use this site to design primers.
    • Operon Order DNA oligos here.
  • Literature search.
    • Medline  Search literature for reports of your gene, including lac fusion and knock-out mutants.
    • ASM Journals  Search J. Bacteriology and other ASM journals.  Any article found here we can download (see Dr. S. or Chris).
